Targeted transgenic mice: targeted results

Our targeted transgenic mouse projects now use goGermline, the revolutionary new technology to generate transgenic mice fast and efficiently. The aim of our targeted transgenic service is to generate a single mouse line, where the only genetic modification is the change in the targeted locus. This is not true for random transgenics, where you end up with multiple independent lines.

Ozgene is making continuous improvements on project timelines by stabilising, standardising and refining each individual process using Lean management. Ozgene’s fastest gene targeting project was recently completed in 20 weeks and our current average project timeline is 30 weeks.

  • What do we mean by targeted transgenic mice? >

    Rather than randomly integrating the transgene into the mouse genome, the targeted transgenic mouse approach uses gene targeting by means of homologous recombination in ES cells. The transgene cDNA is knocked into a “safe haven” locus in the mouse genome to generate transgenic mice with a predictable expression pattern.
  • What is our specific method for producing transgenic mice? >

    Ozgene utilises the mouse gene, Gt(ROSA)26Sor, to generate targeted transgenic mice. The genomic structure of this gene, commonly referred to as ROSA26, is conserved between mice and humans. We use this technology since it provides you with the following benefits:

    1. Only 1x transgenic mouse strain is needed. Not multiple founder lines that may or may not express your gene of interest.
    2. This saves time, money, resources, genotyping, phenotyping, and animals.
    3. The 3Rs (refine, reduce, replace) will be achieved for the Institutional Animal Care and Use Committee (IACUC).
    4. Targeting into the ROSA26 locus achieves an ubiquitously inducible expression pattern.
    5. Data is available for most tissues in the form of ROSA lacZ/eGFP reporter mice that have been crossed with many different tissue specific Cre mice.
    6. A predictable transgene expressing animal is produced.
    7. The ROSA targeted strain has more utility, as it can be used for tissue specific transgene expression just by using a different Cre mouse.
    8. Unwanted side effects caused by insertional mutagenesis associated with random transgenic mice are avoided.
    9. The targeted transgenic mice are readily bred to homozygozity.
    10. You can directly compare data between targeted transgenic mouse lines. For example, various different point mutations can be compared in independent transgenic mice that you made or have been published.
  • What type of research benefits from targeted transgenic mice? >

    Our targeted transgenetic mice are very useful in preclinical drug validation. These mice enable you to precisely model human disease with subtle manipulation of the mouse genome.

While Ozgene currently generates all transgenics via targeting into the ROSA26 locus, the papers also feature transgenics generated by pronuclear injection and lentiviral transgenics, which were previously offered as a service.
